Can it simply be lifted up to open?

Manually unlocks the transmission or something to that nature. I've seen it mentioned in a few threads where people were stuck in park.

Undoes the P lockout. If you need to be towed, pull that cover off, push in the button and you can take the truck out of Park without the truck running.

Do you know if it can simply and safely just be opened without messing up anything? I have a lot of dirt/debris I wanted to clean out of it.

^^^^yup you popthat thing off and you can manually depress the switch so you can get out of park with out depressing the brake pedal. I think you also use it to take off the center console because you have to more the shifter down iirc

Stick a plastic trim tool or a flat head screw driver with some tape/towel around it in the gap and pry it up. It moves quite easily.

Just pull up the latch and go at it with a vacuum or something and you should be fine cleaning it out.

If you mean remove the center console plastic around the shifter, you don't need to mess with that latch. Just pull up at all 4 corners. The PRND portion just snaps in place as well.

The dealer tried to deliver my truck without it. While signing off paperwork to buy the truck I mentioned that this was missing and that they should fix that. Two days later when I took delivery of the truck, I asked if they fixed it - nope. Got the cover in the mail a week later.

Is there anyway to replace this tab? It’s stuck in the up position and have to manually depress it when I need to get into drive. Any suggestions besides cramming something in there?

It is supposed to be UP.
You should not have to press it Down if your foot is on the brake pedal while trying to shift.

    Are your brake lights working?

Shift lock override button. It is to allow you to shift out of P with a dead battery or faulty shift lock solenoid. The cover does not come installed from the factory but in a little bag tied around the shifter. It is installed during PDI but can often be misplaced especially during dealer trades.

It’s really cheap if it’s missing and can be had from any Toyota dealer.

So, your brake lights weren’t working. Do you have any idea when and why the fuse blew? Towed a trailer lately? Janky wiring on a topper 3rd brake light?

LATE late reply but yes the brake lights were not working and it was from a leaky topper into the 3rd brake light. Ended up driving home for 2 hours and thankfully didn’t cause any wrecks. Hand signals the whole way.

IF you have the same issue… PLEASE CARRY FUSES IN YOUR TRUCK. I was blessed to make it home on snowy roads but not having brake lights without knowing is noooo good. Hopefully some 2nd gen people see this.
